In central project Z1, the Waisman laboratory and Reifenberg’s CLAF will provide advice and support to the other laboratories of the Transregio in generating the targeting vectors for the generation of transgenic mice, for conventional and conditional gene targeting, and for the conditional expression of genes. Manipulate ES cells will be generated in the Waisman laboratory, and injected to produce chimera mice in CLAF. In addition, CLAF will generate (BAC)-transgenic mice by pronuclear injections. The laboratory of Ari Waisman specializes in the generation of different mouse mutants using embryonic stem cells (ESC) and bacterial artificial chromosomes (BACs). In the last years the laboratory was able to generate a variety of gene targeted mice, including conditional gene targeting and `knock-in` mice for the overexpression of different transgenes in a tissue-specific and spatial fashion. Recently, the laboratory was able to use the technique of Red/ET cloning for the generation of BAC-transgenes that allow faithful expression of transgenes, for example the Cre-recombinase. The Central Laboratory Animal Facility (CLAF) of the University of Mainz is headed by Kurt Reifenberg. CLAF is equipped with 2.5 veterinary and about 25 animal technician positions and is responsible for all aspects of the housing of laboratory animals and supervises the animal work of the different scientific groups of the University and the University Clinic. The biotechnology laboratory of CLAF offers a service for the generation of knockout, knock-in or transgenic strains by microinjection of embryonic stem cells (ESC) or DNA into murine embryos, which inter alia shall be substantiated by the current Z1 project. Together, the Waisman laboratory and Reifenberg’s CLAF are able to provide advice and support to the other laboratories of the Transregio in generating the targeting vectors for conditional gene targeting and for conditional expression of genes. The ESC will be generated in the Waisman laboratory, and injected to produce chimera mice in CLAF. In addition, CLAF will generate (BAC)-transgenic mice by pronuclear injections. Further, a new reporter mouse will be generated in the frame of the project Z1, the ROSA26-hCD2-IRES-mRFP1 mouse. This mouse will allow to track Cre-expressing cells (for example after cross to Th1-Cre mice) as these cells will express RFP, and in addition, these mice will allow to sort the Cre positive cells, using the hCD2 surface expression with magnetic beads.
