We here analyze the resistance to apoptosis through cellular redistribution of the pro-apoptotic BCL-2 proteins BAX and BAK in hepatocellular carcinoma (HCC). The distribution of BAX and BAK between cytosol and mitochondria will be analyzed in tumor and corresponding non-tumor samples from 34 HCC patients. Key transcriptome changes and gene expression profiles related to specific BAX localization will be applied to two independent cohorts including over 500 HCC patients. The prediction of apoptotic response will be tested using cell lines and polyclonal tumor isolates and cellular protection from BAX will be confirmed by ectopic expression of mitochondrial BAX.
