Many studies published since the year 2000 establish the importance of retrograde endocannabinoid (eCB) signaling in the regulation of synapses throughout the brain. We have a strong body of published and unpublished preliminary data, suggesting that the CB1 cannabinoid receptor is expressed in sympathetic terminals and transmits retrograde signals that restrain norepinephrin (NE) release. Our studies in osteoblasts further show that isoproterenol (ISO), a ß-adrenergic receptor agonist having NE-like activity, stimulates production of the major eCB, 2-AB. Based on these data we hypothesize that 2-AG and NE function in a negative feedback circuit that regulates the synaptic-like activity between sympathetic terminals and peripheral cells.
