Development of Rat Autoimmune Glaucoma Model - 8 Week Study
Laufzeit: 01.01.2005 - 31.12.2007
Kurzfassung
To investigate if human Heat Shock Proteins (HSP 27, HSP 60) induced immunization will cause retinal ganglion cell (RGC) loss in Lewis rats similar to the RGC loss found in glaucoma patients.
RGCs will be counted in Brn-3 immune-stained retinas to determine the RGC loss in immunized animals in comparison to the healthy control animals.
Optic nerves axons damage will be scored by reading optic nerve cross sections.
Conscious intraocular pressure (IOP) will be measured with TonoLab before...To investigate if human Heat Shock Proteins (HSP 27, HSP 60) induced immunization will cause retinal ganglion cell (RGC) loss in Lewis rats similar to the RGC loss found in glaucoma patients.
RGCs will be counted in Brn-3 immune-stained retinas to determine the RGC loss in immunized animals in comparison to the healthy control animals.
Optic nerves axons damage will be scored by reading optic nerve cross sections.
Conscious intraocular pressure (IOP) will be measured with TonoLab® before immunization and 4 and 7 weeks after immunization.
Fundus pictures will be taken in sedated animals (Ketamine and Xylazine) before immunization and 4 and 7 weeks after immunization.
Blood will be collected from all animals before immunization and 4 and 8 weeks after immunization. The serum samples will be used to analyze changes in the antibody and protein profile.
Aqueous humor will be collected on the day of harvesting eyes.
